According to research by the University of Michigan Life Sciences Institute, blocking the activity of an enzyme in fat cells can reduce obesity and other health problems in mice. The study was published online in January 17 in Nature Metabolism. It focused on an enzyme called histone-deacetylase 6 (or HDAC6) to decrease obesity and improve the brain’s ability detect the hormone leptin. Excess energy in the form fat accumulates in animals and humans. Fat cells release more of this hormone into the body’s circulation. This hormone signals the brain to reduce appetite and increase calorie burn, which helps restore energy balance. “When the role of leptin first became known, people believed it would cure obesity,” stated Isin Cakir, a metabolic researcher at the LSI who was also the lead author of this study. “If leptin is able to lower food intake and increase energy burn, then more of it should be helpful in reducing obesity.” This was not the case. Problem is, as obesity increases, the body becomes less sensitive to leptin. Therefore, even higher levels of circulating Leptin don’t affect appetite or energy expenditure. This is true for humans and mice alike. Cakir and his associates have found a way to make mice sensitive to leptin, which is already produced in their bodies, and to induce weight loss and improve metabolic function. Researchers treated obese mice who had been given a high-fat diet using a compound that inhibited HDAC6. The mice lost almost 25%; their body weight within a few weeks. This was not the usual weight loss associated with caloric restriction. The mice’s overall metabolic health was also significantly improved by the team. They didn’t experience the decreased energy expenditure associated with reduced food intake. In fact, they showed significant improvements in liver health and glucose tolerance. This indicates that they are less likely to develop diabetes. The same compound was not used to treat obese mice. These results confirm that HDAC6 inhibition of obesity must be at high levels. Furthermore, altering HDAC6 activity can increase body weight by increasing the sensitivity to natural Leptin. Although the results are encouraging in mice, Cakir emphasizes that they are not yet able to translate into obesity treatment for humans. He said that there are many compounds that can reduce obesity in mice, but not in humans. Or that may reduce weight in humans, but they aren’t safe. The most important questions are: Will HDAC6 inhibition have the same effect on humans and will it be safe? We still have a lot of research to do before we can answer those questions. The most potent HDAC6 inhibitors can cause toxic reactions in the body if a portion of the molecule is broken down. Although this toxicity may be manageable for certain cancers, it is not possible for those suffering from diabetes and obesity. Cakir and his colleagues developed HDAC inhibitors without the toxic component but with the same activity against HDAC6 rodents. The National Institutes of Health, the American Diabetes Association, American Heart Association, and Canadian Institutes of Health Research supported the research. The study authors include: Cakir, Colleen hadley, Pauline Lining Pan and Qiuyu Wang. McKinsey serves on the scientific advisory boards of Artemis Bio, Eikonizo Therapeutics and received funding from Italfarmaco to fund a separate project. He also has a subcontract with Eikonizo Therapeutics for Small Business Innovation Research grants from the National Institutes of Health. Cakir, White and Lee have filed a provisional application for a patent on non-hydroxymate HDAC6 inhibitors. The Australia Institute has revealed that only a third of gas exports from Western Australia are subject to tax or royalties. Liquefied Natural Gas (LNG) worth $27 billion was exported from WA last year, while only $430m went to the State Government, just 1% of state budget revenue.
